Citrulline Dose for Blood Pressure Studies: 3-9 g Explained
The citrulline dose for blood pressure studies was 3 to 9 grams of l-citrulline a day, taken for 1 to 17 weeks in small groups of adults. A Melonex capsule holds a 500 mg blend shared by three ingredients, so it sits far below those trial amounts, and no trial tested the capsule itself.

No. 01The numbers
What was the citrulline dose for blood pressure studies?
The citrulline dose for blood pressure studies was 3 to 9 g a day of oral l-citrulline, tested in 8 randomized trials.
Each trial was small, with 12 to 34 people, and the volunteers ranged from about 22 to 71 years old. They took citrulline for 1 to 17 weeks (Barkhidarian 2019, citrulline and BP).
On average, systolic pressure was 4.10 mm Hg lower than with placebo. The diastolic difference, 2.08 mm Hg, was not statistically significant (Barkhidarian 2019, citrulline and BP). At 6 g a day or more, the diastolic drop reached 2.75 mm Hg.
Those are averages across groups. They say little about how any one person would respond. A drop of 4 mm Hg is small next to the gap between normal and high pressure. Doctors treat high pressure with proven plans, and a supplement trial is not one.

How does a capsule compare with the trial amounts?
The table sets each label line against the studied range, using the label's order and the 21 CFR 101.36 rule.
The label lists the blend heaviest first, so each place has a ceiling. These are weights of the salt forms, not free amino acids.
| Label line | Place | 1 capsule | 2 capsules (label max) | Amount used in research | Evidence |
|---|---|---|---|---|---|
| L-Citrulline DL-Malate | 1 of 3 | at most 500 mg | at most 1,000 mg | 3-9 g a day of L-citrulline in blood pressure trials (1-17 weeks) (Barkhidarian 2019, citrulline and BP) (NIH ODS, arginine and citrulline) (RxList, L-citrulline) | Small systolic drop in short, small trials; no trial of Melonex |
| L-Arginine (Alpha-Ketoglutarate dihydrate) | 2 of 3 | at most 250 mg | at most 500 mg | 4-24 g a day of L-arginine in blood pressure trials (Dong 2011, arginine and BP) (Mayo Clinic, L-arginine) (RxList, alpha-ketoglutarate) | Small drops in blood pressure in trials of grams a day; no trial of Melonex |
| L-Arginine HCL | 3 of 3 | at most 167 mg | at most 333 mg | 4-24 g a day of L-arginine in blood pressure trials (Dong 2011, arginine and BP) (Mayo Clinic, L-arginine) (VINTAGE MI trial (ACC summary)) | Same arginine research; the salt form adds no evidence |
| Hydroxypropyl Methylcellulose (capsule) | Other ingredient | not listed (inactive) | not listed (inactive) | Not a nutrient; it forms the capsule | Inactive ingredient |
| Microcrystalline Cellulose | Other ingredient | not listed (inactive) | not listed (inactive) | Not a nutrient; a filler | Inactive ingredient |
| Magnesium Stearate | Other ingredient | not listed (inactive) | not listed (inactive) | Not a nutrient; a flow agent | Inactive ingredient |
The trial range is grams, and the capsule's top figure is hundreds of milligrams. No row in the table is a measured amount. Use it to see scale, nothing more.
No. 03Worked example
How many capsules would match 3 grams of citrulline?
On paper, about 11 capsules at the very most, which is far above the label's limit of two, so the label and the trials are not meant to line up.
Take the best case first. If the whole 500 mg were citrulline malate, one capsule holds about 283 mg of citrulline, using the NIH ratio of 56.64% (NIH ODS, arginine and citrulline). Divide 3,000 mg by 283 mg and you get roughly 10.6 capsules.
Now take an even split. Each ingredient is then about 167 mg, giving only about 95 mg of citrulline, and 3 g would need over 30 capsules.
This is arithmetic, not advice. The label says: "Do not exceed recommended dose." It allows one capsule a day, and up to two.
No. 04Safety
Is higher-dose citrulline known to be safe?
Short studies saw no adverse effects up to 6 g a day for 4 weeks, but the NIH says citrulline's safety has not been adequately assessed (NIH ODS, arginine and citrulline).
Those trials were short, and the people in them were few. Nothing in our sources covers years of use. Think of it as a short walk, not a long road.
- Pressure drugs may stack with citrulline, and pressure may go too low (RxList, L-citrulline).
- Nitrates and erectile dysfunction pills are listed as major interactions (RxList, L-citrulline).
- Drug references advise avoiding it in pregnancy and nursing for lack of data (RxList, L-citrulline).

No. 05What it means
So what should you take from the dose gap?
Take the gap as a reason to keep expectations modest and your doctor in the loop.
The research is interesting, but it is about grams of one form, taken under study conditions. The capsule is a different thing, so the studies cannot be borrowed for it. The benefits page sets the label wording beside the evidence.

Melonex is a dietary supplement, not a treatment. Ask your doctor or pharmacist first if you take a nitrate for chest pain, an erectile-dysfunction drug such as sildenafil, any blood pressure medicine, a potassium-sparing diuretic, a blood thinner or a diabetes medicine, or if you have had a heart attack. Arginine and citrulline can push blood pressure down further. Keep taking prescribed medicine. See side effects and interactions.

Melonex questions people ask
How many grams of citrulline did blood pressure trials use?
Trials of oral l-citrulline used 3 to 9 grams a day for 1 to 17 weeks, in small groups of adults. That is the range in the 2019 meta-analysis we cite. No trial tested a Melonex capsule.
Is 500 mg of citrulline enough to matter?
We cannot say. Trials used 3 to 9 g, and a 500 mg blend holds at most about 283 mg of citrulline even if all of it were the malate salt. Research on that lower amount for blood pressure was not found in our sources.
Did the studies include people on blood pressure drugs?
The abstract we read does not break that out, so we cannot say. What is known is that citrulline may push pressure too low beside pressure drugs. Ask your doctor or pharmacist before combining them.
